PT-141 dosing window - how long before activity for optimal effect

Been using melanotan variants for years. Know the mechanism, MC4R agonism, central not peripheral. Not asking about that.

What I actually can't nail down is the timing. Every source says something different. 30 minutes, 2 hours, 4 hours. The older MT-II literature suggested fairly rapid onset but PT-141 seems to behave differently in practice even though the mechanism is similar.

Running 1mg intranasal currently. Effects are real but timing feels inconsistent. Some sessions the response is almost immediate, other times it's sluggish for over an hour then hits hard.

Is this a variable absorption issue with intranasal vs subQ, or is there something about MC4R receptor saturation state that affects onset time? Wondering if dosing consistency matters here, meaning does receptor downregulation between doses affect the timing or just the magnitude.

Not interested in anecdotal "it worked great for me at X time" responses. Looking for anyone who's done systematic timing comparisons or knows the pharmacokinetic literature well enough to actually address this.

Ok so I have to ask because this is something I've been trying to figure out too and your experience with the inconsistency really resonates with me...

I've noticed the exact same thing and it honestly drove me a little crazy for a while! My question for you is - have you tracked what's different between those sessions? Like I started keeping a little notes app log and noticed that for me the faster onset days were almost always when I was more hydrated and hadn't eaten a big meal recently. The slower onset days seemed to cluster around times when I was tired or had eaten heavy beforehand.

I know you said you're not looking for anecdotal stuff and I totally respect that, but I'm genuinely curious whether you've ruled out those physiological state variables before going to the receptor saturation explanation. Because I wonder if some of that inconsistency might actually be downstream of pretty mundane stuff rather than anything happening at the MC4R level specifically.

Also are you timing from dose to actual onset, or dose to peak effect? Because for me those feel like pretty different windows and I wasn't always clear which one I was measuring at first which made my self-tracking kind of a mess for a while lol.

What's your dosing frequency between sessions?
